Design Notes
This RNA aptamer has not been tested in the lab by our team, however the Edinburgh iGEM 2020 team tested the BBa_K3380150 RNA aptamer in the BBa_K3380500 construct and BBa_K3380600. Similar design can be used for the DIR2s-Apt RNA aptamer.
All the sequences for the individual parts were acquired. The individual part were designed with 4bp scars to allow swapping of the parts to test different constructs. The individual parts were phosphorylated, annealed and ligated together using the T4 ligase.

The DIR2s-Apt is an artificial RNA aptamer obtained by testing its binding properties to the DIR (dimethylindole red) fluorophore.
